It is available as a dietary supplement and cream resveratrol.
For years, reports on what to do pharmacological resveratrol were conducted with knowledge of the mechanisms of absorption and bioavailability of this compound. When you radiolabeled tracer trans-resveratrol helped make the first observations, the surprise was great to see that shortly after an oral dose, it can be found primarily in the urine as conjugated and n types that was barely detectable in the blood plasma. Resveratrol antioxidant is absorbed orally ingested up 70% in the gut, which is a remarkably high rate for a [13] polyphenols. It appears that in some subjects quickly (30 my) after ingestion of traces antioxidant while in the blood plasma, but is rapidly metabolized in the liver of resveratrol-3-sulfate (or -4 ‘-sulfate) or resveratrol -3 four “-disulfate(or 3,5-disulfate) and resveratrol-3-O-glucuronide (or 4′-O-glucuronide).Glucuronidation and 4 “seems to preferentially [14].En 2003, Goldberg [15] deduce that, “it seems that the position of welfare of the positive aspects of these compounds in their genre unconjugated, mainly based on scientific studies of the in vitro activity, was unrealistic and has been greatly exaggerated.”
Research in the distribution of 14C-trans-resveratrol antioxidant in all tissues of the mouse can be completed very accurately by autoradiography. Vitrac et al (2003) [16] found that the emphasis antioxidant of radioactivity in blood was relatively low antioxidant and continues through 6 h in the experiment. After 3 h, we find the greatest concentration in the duodenum, and after this order, kidneys descending, the liver and lungs, spleen and colon, the heart, the brain and testes. Use antioxidant microautoradiographie, the authors have shown that trans-resveratrol can penetrate tissue, especially the liver hepatocytes where exactly antioxidant are capable of incorporating radioactivity. Given the presence of resveratrol glucosides (picéide, astringine) in red wine and their possible hydrolysis within the gut, the authors conclude: "It may be proposed antioxidant that the reasonable consumption of red wine and typical human body must ensure sufficient quantity of resveratrol for action against cancer chemoprevention.
Lipid degradation by oxidative stress cause deposits of oxidized lipids in the vessels at the origin of atherosclerotic plaques, disrupt the functioning of cell membranes and produce carcinogenic derivatives of resveratrol.
Radical attacks the DNA will be a source of carcinogenic mutations and those proteins inhibit enzymes and disrupt the cell signaling of proliferation or defense.
Antioxidants can act at two levels of oxidation reaction :
- from the initiation phase, they can prevent radical formation by blocking, in complexes, transition metals (iron, Copper) otherwise acting as powerful catalysts. These are antioxidant chelating transition metals.- during the propagation phase, they can intercept radicals and break the chain reaction. They are scavengers radicaux.Le resveratrol, like all the polyphenols, has phenolic hydroxyl groups, Ar-OH, H can provide the peroxy radicals and thus, neutralize them as hydroxides L-OOH :
Resveratrol is a antioxidant that helps keep healthy longer
Since there is no absolute measure for assessing the potential antioxidant a compound, should be used for comparative assessments with other compounds. Moreover, these comparisons will depend on the assessment method and especially the free radical peroxyl antioxidant used.
By comparing the in vitro ability to scavenge free radicals (2,2-diphényl 1-picrylhydrazyl) by resveratrol and flavonoids in wine, Iacopini and its collaborators have obtained the following hierarchy : Resvératrol.Les flavonoids are antioxidants than vitamin E (Trolox) while resveratrol is much moins.Mais if using a generator of radicals involved in pathophysiological processes such as peroxynititre , we obtain the following order: Resveratrol is then in the average potential antioxidant flavonoids.
A test even more interesting in terms of assessing the pathophysiological ability to reduce the peroxidation of low density lipoprotein LDL (carrier of cholesterol). It is now widely accepted that the formation of free radicals from lipid antioxidant LDL is a key factor in the genesis of atherosclerosis. By incubating LDL with cupric ion Cu 2 for catalytic, the effect of inhibition of lipid peroxidation, obtained by adding polyphenols, is in the following order : CatécholLe resveratrol is a powerful antioxidant, even more effective than flavonoids prevent LDL peroxidation. It acts primarily "upstream" of the reaction by making irrelevant the catalyst cupric antioxidant. It is a chelator of copper ion Cu2 . The great power of chelation of resveratrol in vivo is useful because it is known that LDL has a high propensity to bind copper antioxidant. The chelating ability of cis-resveratrol antioxidant by cons is only half that of the trans-resvératrol.Lorsque LDL is incubated with as free radical generator, l’AAPH, resveratrol acts "downstream" of the reaction by trapping free radicals formed from lipid peroxyl and preventing the spread of the chain reaction of oxidation. Resveratrol, however, is a scavenger of free radicals less potent than flavonoids antioxidant and roughly similar to vitamin E : Trolox. With another technique designed to highlight the mechanism antioxidant scavenger of free radicals (and not chelating), Sojanovic et al (2001) showed that resveratrol and flavonoids antioxidant, quercetol and epicatechin, were all equally powerful. All three were able to reduce approximately 95% lipid peroxidation liposomes.L effect antioxidant Wine should be attributed to the flavonoids and phenolic acids, rather than resveratrol, since the latter is present at levels several hundred times lower (0,1 – 10 mg· trans-resveratrol cons 1000 – 3 – mg·flmgonoids). They also showed that it was mainly the hydroxyl group OH H gave his position to trap free radicalsantioxidant.
